The method for solving the problem of high column pressure in high performance liquid chromatography
When the HPLC is turned on or the column is changed, the HPLC column pressure is too high. This is a common problem. There are two reasons for the column pressure being too high: 1. The reason for the column itself. Mainly manifested in long-term use caused by sample contamination caused by blockage, causing excessive column pressure. Or the column is loosened by physical vibration during packing or transportation, etc. The problem of the column itself. Second, the column pressure is too high is usually not a chromatogram. The problem of the column itself is the chromatographic instrument or the liquid pipeline, and the flow is caused by external causes. When the column pressure is too high, what should we solve? First, we should find out the cause of the column pressure too high, the main steps There are:
1. Remove the pre-column to see if the column pressure is still high, otherwise it is the problem of protecting the column. If the column pressure is still high, check again;
2. Remove the column from the instrument to see if the pressure drops. Otherwise, the pipeline is clogged and needs to be cleaned. If the pressure drops, check again;
3. The inlet and outlet of the column are reversed to the instrument, and the column is washed with 10 column volumes of mobile phase (at this time, do not connect the detector to prevent solid particles from entering the flow cell). At this time, if the column pressure still does not drop, check again;
4. Replace the column inlet screen. If the column pressure drops, your solvent or sample contains particulate impurities. It is these impurities that block the screen and cause pressure rise.
